Summary

Gram-positive bacteria share an important cell-wall feature, but they are not all alike. This lesson introduces the Gram stain and the molecular evidence used to distinguish major groups.

A typical gram-positive cell wall contains a thick, multilayered matrix of peptidoglycan. During the Gram stain procedure, the primary stain, crystal violet, is retained and stabilized in this matrix. As a result, these cells usually appear purple when viewed with a brightfield microscope.

All flashcards

Why are mycobacteria visualized with an acid-fast stain?
Their mycolic-acid-rich waxy coat restricts penetration by Gram stain reagents.
How does botulinum neurotoxin cause flaccid paralysis?
It blocks acetylcholine release at neuromuscular junctions.
What are Lancefield groups based on?
Shared antigens used to classify streptococci.
Why does Mycoplasma not Gram stain?
It lacks a cell wall, so Gram stain reagents do not stain it.
What is bioprospecting?
The search for useful biological resources, such as microorganisms or their products.

Quiz questions

  1. What is the main survival advantage of bacterial endospores?Answer: They are dormant, resistant structures that can persist through unfavorable conditions.
  2. How does the neurotoxin from Clostridium tetani produce its characteristic paralysis?Answer: It interferes with inhibitory signaling in the central nervous system.
  3. Which description fits Streptococcus pyogenes?Answer: A group A, beta-hemolytic streptococcus associated with pus-forming infections.
  4. How can Arthrobacter's cell shape change with growth phase?Answer: It may be rod-shaped during exponential growth and coccoid in stationary phase.
  5. What do metachromatic granules in some Corynebacterium cells store?Answer: Inorganic phosphate.
